Title / Titel Microsatellites in the water frog model system

Summary / Zusammenfassung Microsatellites are short DNA sequences of about 2-4 nucleotides that occur in multiple repeats of varying numbers of the unit. They are analyzed via PCR, using primer pairs designed from DNA sequences flanking the repeat regions. As genetic markers, micro-satellites are highly informative (single loci; codominant alleles; hypervariable but genealogically sufficiently stable; obtainable from small amounts of tissue). We are developing microsatellite loci for the water frog group, in collaboration with Thomas Uzzell (Philadelphia) and others, and so far have cloned 500 independent loci that hybridized to a mixture of tri- or of dinucloetide repeats. We have sequenced a subset of > 100 of these clones; 80% of consensus insert sequences containted uninterrupted direct repeats of at least 6 units. We have designed primer pairs for 19 of these sequences and are currently testing for amplifiability and for variation within and between populations and species.

Microsatellite markers will be used for (1) studies of Swiss populations (variation among hybrid Rana esculenta lineages, colonization history of Rana lessonae and Rana esculenta, amount of inbreeding, degree of isolation between populations, dispersal rates); (2) studies of colonization history of species from refugia of the last glaciation period to their present range and the origin of hybrid lineages with clonal reproduction; and (3) the analysis of hybrid zones between genetically divergent populations in southeastern Europe.
