| Summary / Zusammenfassung | Clustering of acetylcholine receptors (AChRs) in the muscle membrane is one of the first signs of postsynaptic differentiation during formation of the neuromuscular junction and can be induced by components of the basal lamina such as agrin and laminin. We analyze how other postsynaptic proteins are redistributed by these two components, how they interact with AChRs to cause clustering, and what role AChRs themselves play in this process. The current data indicate that laminin and agrin act by different yet related pathways, as some postsynaptic proteins are differentially distributed after treatment of muscle cells with either factor, whereas tyrosine kinases and the scaffolding protein rapsyn are required in both cases. Furthermore, muscle cells were found to contain pre-assembled complexes of AChRs in association with other postsynaptic proteins, and such complexes play a role in postsynaptic assembly induced by agrin.
